搖床
搖床(chuang)(chuang)又稱搖瓶機,它是培(pei)養好氣性微生物的小型試驗設備或(huo)作為種子(zi)擴大(da)培(pei)養之用(yong),常用(yong)的搖床(chuang)(chuang)有往復(fu)式(shi)和旋轉式(shi)兩種。往復(fu)式(shi)搖床(chuang)(chuang)的往復(fu)頻(pin)率一般(ban)在80~140次(ci)(ci)/min,沖(chong)程(cheng)一般(ban)為5~14cm,如頻(pin)率過快、沖(chong)程(cheng)過大(da)或(huo)瓶內液(ye)(ye)體(ti)裝量過多,在搖動(dong)(dong)時液(ye)(ye)體(ti)會濺(jian)到(dao)包(bao)扎瓶口(kou)的紗布或(huo)棉塞上(shang),導(dao)致雜菌污染,特別(bie)是啟動(dong)(dong)時更容易發生這種情況。旋轉式(shi)搖床(chuang)(chuang)的偏心距一般(ban)在3~6cm之間(jian),旋轉次(ci)(ci)數為60~300rpm。

液體(ti)(ti)菌(jun)(jun)種(zhong)接種(zhong)箱(xiang)是專為移(yi)接液體(ti)(ti)菌(jun)(jun)種(zhong)而(er)設(she)(she)(she)計的。比固體(ti)(ti)菌(jun)(jun)種(zhong)箱(xiang)窄長,單側兩(liang)人操(cao)作。內設(she)(she)(she)軌道和紫外線燈(deng),箱(xiang)兩(liang)端開有高(gao)25cm,寬(kuan)10cm的長方(fang)形出(chu)(chu)口(kou)(kou),方(fang)便菌(jun)(jun)種(zhong)進出(chu)(chu),洞口(kou)(kou)設(she)(she)(she)有小推門。進出(chu)(chu)口(kou)(kou)下處設(she)(she)(she)蒸汽源,接種(zhong)時用(yong)蒸汽封(feng)住進出(chu)(chu)口(kou)(kou),以防雜菌(jun)(jun)進入(ru)箱(xiang)內。箱(xiang)背面設(she)(she)(she)有液體(ti)(ti)菌(jun)(jun)種(zhong)移(yi)接管能進入(ru)的小孔。
接(jie)(jie)(jie)種箱滅(mie)菌(jun)時(shi)(shi),用紫(zi)(zi)外線照(zhao)射30min。如果沒(mei)有紫(zi)(zi)外線燈,可用甲醛和高錳酸(suan)鉀(jia)(甲醛10~14mL/m3+高錳酸(suan)鉀(jia)5~7g/m3空間)熏蒸30min以上。使用時(shi)(shi),先將所需物(wu)品和工具放(fang)入接(jie)(jie)(jie)種箱內,然后進(jin)(jin)行(xing)(xing)藥劑熏蒸和紫(zi)(zi)外線滅(mie)菌(jun),再(zai)按無菌(jun)操作(zuo)(zuo)進(jin)(jin)行(xing)(xing)接(jie)(jie)(jie)種。接(jie)(jie)(jie)種箱的結構(gou)簡單,造價低廉,易消毒滅(mie)菌(jun),操作(zuo)(zuo)方便,而且(qie)人在箱外操作(zuo)(zuo),氣溫(wen)較高時(shi)(shi)也能作(zuo)(zuo)業(ye)。缺點是(shi)進(jin)(jin)出培養基費(fei)工費(fei)時(shi)(shi),每次接(jie)(jie)(jie)種前都需要(yao)進(jin)(jin)行(xing)(xing)滅(mie)菌(jun)。
保鮮柜 微生物實驗室的冰箱主要有兩種:普通冰箱和低溫冷凍冰箱。普通冰箱一般都具有兩個柜子,即鮮藏柜和冷藏柜,溫度分別為4℃和-20℃;低溫冷凍冰箱溫度一般控制在-40~-80℃。它們都可以用于微生物菌種保藏。鮮藏柜常用于保存斜面菌種,保藏時間在3個月左右。超過3個月,斜面就會變干,因此需要轉接菌種。如果要長時間保存菌種,則需要經過處理后,貯藏于普通冰箱的冷藏柜或低溫冷凍冰箱中,它們的保藏時間較長,一般都在1年以上。
